1. Who would make a better college professor: 6 (Michonne) or 11 (Natasha Romanov)?
Tough to say. Both women would be competent at it, definitely, being intelligent and strong-willed women. But I think Michonne, who was canonically a lawyer before the zombie apocalypse, would enjoy it more for itself; whereas for Natasha her reason for being there would probably be a mission or cover? So I would probably enjoy Michonne's classes more.
2. Do you think 2 (John McClane) is hot?
Depends on which era you choose. He's a little past it by the time of "A Good Day to Die Hard". But he fit right into the bald-is-sexy niche in my personal taste in the fourth one, "Live Free or Die Hard" - for which niche I entirely blame a childhood growing up watching ST:TNG. :)
3. 12 (Nick Burkhardt) sends 8 (John Wick) on a mission. What is it, and does it succeed?
Hoo, boy. I can only imagine that it would be some Grimm-related mission to terminate an extremely dangerous wesen that Nick himself could not take on for some reason. And yes, of course; if John Wick took the mission in the first place, it's as good as done.
4. What is, or would be, 9's (Eve Baird's) favorite book?
I can't remember off the top of my head if we heard about her reading preferences in canon. Aside, of course, from the magical mission-giving scrapbook. If that doesn't count, though, and canon doesn't specify - I imagine it would be whatever book Flynn deduced she'd like, and pointed out to her; he is The Librarian after all!
5. Would it make more sense for 2 (John McClane) to swear fealty to 6 (Michonne), or the other way around?
I can't imagine either actually swearing fealty to anyone, other than Lady Justice, perhaps. Though given the previous meme, now there's a plotbunny in the back of my head muttering about Michonne ending up as the queen of some post-ZA society instead of ever running into Rick's group, with McClane as her right hand ....
6. For some reason, 5 (Abbie Mills) is looking for a room-mate. Should they share a studio apartment with 9 (Eve Baird), or 10 (Natasha Romanov)?
Either one is a professional woman who would be gone more than they were around and understand the keeping of secrets, so neither would be a bad choice. Also, at some point during each of their canon, they are all women working with (and attracted to a member of) a small team with whom they save the world. But I think in terms what might happen should they trip over one another's work, the better choice might be Eve, who has no current connections to a large secret organization of controlling habits and dubious morality.
7. 2 (John McClane), 7 (John Reese), and 12 (Nick Burkhardt) have dinner together. Where do they go, and what do they discuss?
Hah. Given Reese's current cover, he and McClane would both be NYPD cops, and Nick probably visiting for a Case of the Weird - so they'd probably end up talking about wackiest/strangest cases they've ever investigated, suitably edited to elide such details as The Machine and wesen. But because his history doesn't need such editing, McClane would likely end up with bragging rights. :)
8. 3 (Liv Moore) challenges 10 (Natasha Romanov) to a duel. What happens?
Depends on whether Liv goes full zombie or not, and whether either one is allowed weapons. Though even if she does, and no weapons are allowed - Natasha is used to fighting bigger and stronger than even a zombie on the hunt for brains. So I suppose she would win, regardless.
9. If 1 (Brian O'Conner) stole 8's (John Wick's) most precious possession, how would they get it back?
Oh, Lord. That's basically the plot of John Wick, right there; so you have only to look to canon to see how that would play out. Much as I love Brian, he wouldn't stand a chance.
10. Suggest a title for a story in which 7 (John Reese) and 12 (Nick Burkhardt) both attain what they most desire.
Canon suggests that it isn't possible for either man to attain what he most desires. But I suppose if I were to attempt it ... "A Time to Kill and a Time To Heal."
11. What kind of plot device would you use if you wanted 4 (Jupiter Jones) and 1 (Brian O'Conner) to work together?
Perhaps he turns out to be another Recurrence of an Entitled, in a family in the same business as Jupiter's (which is why he and Mia and the kiddos won't be in any movie post-Furious Seven)! He goes to take up the reins of what his previous incarnation left him, discovers the whole People Juice thing, and he and Jupiter team up to overthrow the current galactic status quo.
12. If 7 (John Reese) visited you for the weekend, how would you get along?
He'd be quiet, and probably absent a lot on whatever mission brought him there. Since I presume I would be previously acquainted with him from some other job or mission if he was visiting me and not just taking over my property or something, I probably wouldn't expect anything more out of him than that. Maybe cook him a few home meals, make sure the guest bed was freshly made up, and not ask questions about the suspicious case he brought with? Make a social invitation or two that I would be pleasantly surprised if he took up? Etc.
13. If you could command 3 (Liv Moore) to perform any one task or service for you, what would it be?
She's a medical examiner, part-time psychic via dead people's memories, and full-time zombie. There is nothing I can think of that would utilize her skill set. Except maybe have a long conversation about exactly what it's like to have such a weird life, to better inform my future writing? :)

Depends on the threat du jour. Whatever would let him save the most people, because it would have to happen in the midst of some alien or supernatural crisis. He'd have a hard time believing either one, but presented with evidence of people getting hurt, he'd leap right in to stop it.
But trying to imagine them being on opposite sides in the first place ... I suppose it's conceivable that Abbie & Co might go after, say, Caine or Stinger on the basis of their not being human? Especially if someone was misleading them. But it would all come out all right in the end.

Possibly thrown into a place for the criminally insane like her mom and sister if the wrong person overhears her talking about the reasons for it and doesn't buy the cover story!

She knew nothing about swordwork before the apocalypse; the only one she ever wielded beforehand was the novelty letter opener shaped like a dull brass cutlass she kept in her office.
21. If 11 (Natasha Romanov) and 9 (Eve Baird) were racing to a destination, who would get there first?
Depends on whether Eve was allowed to use the Library's Back Door to basically teleport to her destination.
If yes, she'd win; otherwise, Natasha would, with her skills and access to SHIELD/Avengers resources/flying teammates.
22. If you had to walk home through a bad neighborhood late at night, would you feel safer in the company of 7 (John Reese) or 8 (John Wick)?
Let me see, two tall, quiet and deadly attractive men ... *sigh* Probably Reese in the end, as he's a bit more solidly built and distinguished looking, is used to working with a team, and is slightly less likely to leave difficult-to-explain corpses in our wake.
